چکیده

Complexes of the Bowman-Birk soybean inhibitor with either trypsin or chymotrypsin and with both of these enzymes were isolated by gel atration. Amino acid analyses and molecular weight estimates by gel filtration were consistent with the formation of a 1: 1 molar complex between inhibitor and trypsin or chymotrypsin and a 1: 1: 1 molar complex composed of inhibitor, trypsin, and chymotrypsin. The binary complexes were also isolated by aflinity chromatography employing insoluble derivatives of trypsin and chymotrypsin. Determination of the residual enzymic activity in solutions of the isolated complexes permitted an evaluation of their dissociation constants.

Immunoassays for Bowman–Birk and Kunitz Soybean Trypsin Inhibitors in Infant Formula

Because the consumption of soybean inhibitors of digestive enzymes in processed foods may have both beneficial and adverse health-related effects, reliable and rapid analytical methods for these inhibitors are needed. Monoclonal antibody–based sandwich enzyme-linked immunosorbent assays (ELISAs) were developed for the 2 major soybean protease inhibitors, the Kunitz trypsin inhibitor (KTI) and B...

Bowman-Birk Protease Inhibitor as a Potential Oral Therapy for Multiple Sclerosis

The soybean-derived Bowman-Birk protease inhibitor (BBI) is a small protein consisting of 71 amino acids and 7 disulfide bonds [4]. BBI is a double-headed serine protease inhibitor, with two functional active sites at opposite sides of the molecule, which inhibits both trypsin and chymotrypsin-like proteases [1,3] (Figure 1). It is a water-soluble protein that is resistant to acidic conditions ...

Isolation of Bowman-Birk-Inhibitor from soybean extracts using novel peptide probes and high gradient magnetic separation.

Soybean proteins offer exceptional promise in the area of cancer prevention and treatment. Specifically, Bowman-Birk Inhibitor (BBI) has the ability to suppress carcinogenesis in vivo, which has been attributed to BBI's inhibition of serine protease (trypsin and chymotrypsin) activity. The lack of molecular probes for the isolation of this protein has made it difficult to work with, limiting it...

Single-dose administration of Bowman-Birk inhibitor concentrate in patients with oral leukoplakia.

The Bowman-Birk inhibitor (BBI) is a soybean-derived serine protease inhibitor and a potential cancer chemopreventive agent for humans. In this Phase I clinical trial, BBI concentrate was administered as a single oral dose to 24 subjects with oral leukoplakia. Pharmacokinetics of BBI was analyzed, and subjects were monitored clinically for toxic effects. Subjects received between 25 and 800 chy...

Crystal structure of cancer chemopreventive Bowman-Birk inhibitor in ternary complex with bovine trypsin at 2.3 A resolution. Structural basis of Janus-faced serine protease inhibitor specificity.

Understanding molecular recognition on a structural basis is an objective with broad academic and applied significance. In the complexes of serine proteases and their proteinaceous inhibitors, recognition is governed mainly by residue P1 in accord with primary serine protease specificity. The bifunctional soybean Bowman-Birk inhibitor (sBBI) should, therefore, interact at LysI16 (subdomain 1) w...
